The Cutaneous Reaction to the Mixture of Inflammatory Exudate and Bovine gamma-Globulin in Rabbit

Abstract
This study was carried out to confirm the presence of certain factor in inflammatory exudate that mrkcs non-toxic high molecular substance such as bovine gamma-globulin injurious to tissues. Various materials for test were prepared from an inflammatory exudate by some physicochemical methods. The exudate was obtained from the peritoneal cavity of rabbit inflamed by croton oil. Each material alone and together with bovine gamma-globulin was injected intradermally into the abdominal skin in normal rabbits. The cutaneous reaction to various materials injected was observed grossly at definite time intervals for 48 hours and some of the skin lesions were examined histologically.
The results thus obtained were summarized as follows:
1. The inflammation-inducing effect of the mixture of the exudate and bovine gamma-globulin was more striking than the exudate alone.
2.The inflammation-inducing effect of the mixture of the exudate heated at 100¡ÆC for 15 or 30 minutes and bovine gamma-globulin was as striking as that of the mixture of the exudate and bovine gamma-globulin, but the mixture of the exudate heated at 100¡ÆC for 60 minutes and bovine gammaglobulin was less striking.
3. The inflammation-inducing effect of the mixture of chloroform-insoluble substance and acetone-or trichloracetic acid non-precipitating substance of the exudate and bovine gamma-globulin were more striking than that of each substance alone.
4. The inflammation-inducing effect of the mixture of chloroform-soluble substance and acetone-or trichloracetic acid-precipitating substance of the exudate and bovine gamma-globulin was similar to that of each substance alone.
On the basis of the above, it is suggested that in inflammatory exudate there may be not only active ch mical mediator that causes inflammation, but there may be certain factor that makes high molecular substance such as bovine gamma-globulin injurious to tissues, and that the former is heat-labile, nonlipid proteinaceous substance and the latter is heat-stable, water-soluble, non-lipid, non-proteinaceous substance.
